What is Triptorelin?
Triptorelin is a synthetic peptide that mimics a natural hormone called gonadotropin-releasing hormone (GnRH). You may also see it listed under brand names like Trelstar or Decapeptyl, or as its salt form, triptorelin acetate (CAS 140194-24-7).[5] It belongs to a class of compounds called GnRH agonists — drugs that act like GnRH but with a much stronger, longer-lasting effect. The end result? The body's production of sex hormones — testosterone in men, estrogen in women — gets dramatically reduced.[1] That hormone-suppressing ability is what makes triptorelin so interesting to researchers across several medical fields.
How Triptorelin Works
Think of your brain's hormone system like a light switch. Normally, your hypothalamus (a tiny brain region) sends short pulses of GnRH to the pituitary gland, which responds by releasing hormones that tell the gonads (testes or ovaries) to make sex hormones. Triptorelin is like holding that light switch permanently on. At first, hormone levels spike. But the pituitary quickly becomes overwhelmed and stops responding. The result is a sharp drop in sex hormone production — a state researchers call medical castration.[1] This is completely different from what happens with short, natural pulses of GnRH. The constant stimulation is what paradoxically shuts the system down.
What the Research Shows
Most of the published research on triptorelin focuses on its hormone-suppressing effects in humans and its ovulation-triggering effects in animals.
- Liver safety profile: A clinical review from the National Institute of Diabetes and Digestive and Kidney Diseases found that triptorelin is linked to only a low rate of transient enzyme elevations in the liver during therapy, and has not been convincingly connected to cases of serious, clinically apparent liver injury.[1]
- Endometriosis connection: Research published in The Lancet explored the relationship between GnRH-based hormonal suppression and endometriosis management, highlighting how reducing estrogen levels is central to controlling this condition.[4]
- Veterinary ovulation studies: In equine research, a single 0.1 mg subcutaneous injection of triptorelin acetate in saline solution successfully induced ovulation in mares at a rate of 84.7% — comparable to the traditional gold-standard trigger, hCG (87%).[2] Researchers concluded that triptorelin is a strong alternative in countries where other GnRH formulations aren't available.[2]
- Donkey reproduction: A 2024 study from the University of Pisa tested triptorelin in jennies (female donkeys), which are considered an endangered species in several northern hemisphere breeds. A 0.1 mg subcutaneous dose resulted in ovulation in 11 out of 12 cycles, with an 83.3% ovulation rate between 24 and 48 hours post-injection — significantly better than untreated controls (16.7%).[6]
- Swine estrus synchronization: Japan's Food Safety Commission assessed triptorelin acetate for use in weaned sows to synchronize estrus cycles for fixed-time artificial insemination. They found negligible oral bioavailability (it breaks down in the digestive tract) and no genotoxicity relevant to human health, concluding no acceptable daily intake needed to be established for dietary exposure.[5]
What Triptorelin Is Being Studied For
In research settings, triptorelin is primarily studied in the context of conditions where lowering sex hormones is the goal:[1]
- Advanced and locally advanced prostate cancer — testosterone fuels prostate cancer growth; suppressing it is a core treatment strategy
- Endometriosis — a painful condition where tissue similar to the uterine lining grows outside the uterus; estrogen reduction helps control it[4]
- Central precocious puberty in children — a condition where puberty begins abnormally early; GnRH agonists can pause it
- Veterinary reproductive science — ovulation induction in mares[2] and jennies[6], and estrus synchronization in sows[5]

Mixing and Storing Triptorelin
In research settings, triptorelin is typically supplied as a lyophilized powder (freeze-dried) that must be reconstituted before use. The veterinary studies above used triptorelin dissolved directly in saline solution for subcutaneous injection.[2] General research-lab principles apply: use sterile bacteriostatic water or saline for reconstitution, mix gently (swirl — don't shake), and allow the powder to fully dissolve before use. Once reconstituted, the solution should be stored refrigerated (2–8°C) and used promptly, as peptides can degrade. Unused lyophilized powder should be kept sealed, away from light and moisture, and stored at the temperature specified by the supplier.
